MYC(8q24)基因探針
myc基因是較早發(fā)現(xiàn)的一組癌基因,包括C - myc,N -myc,L - myc ,分別定位于8號(hào)染色體,2號(hào)染色體和1號(hào)染色體。結(jié)構(gòu)上由不編碼蛋白質(zhì)的第1外顯子和編碼蛋白質(zhì)的第2、3外顯子構(gòu)成,與之同源的病毒癌基因存在于MC29及其它一些具有高度致癌性的猿逆轉(zhuǎn)錄病毒中。myc基因高水平表達(dá)時(shí)可轉(zhuǎn)化嚙齒類成纖維細(xì)胞。
使用細(xì)胞遺傳學(xué)方法、Southern blot、PCR技術(shù)檢測(cè)c-MYC基因斷裂重組都有其局限性。細(xì)胞遺傳學(xué)方法由于樣本有絲分裂的低比率以及中期分裂相的質(zhì)量問題導(dǎo)致部分樣本檢測(cè)受限;Southern blot、PCR檢測(cè)c-MYC基因相關(guān)的易位需要多種針對(duì)性的引物,耗時(shí)且不利于實(shí)際操作。用FISH探針檢測(cè)c-MYC基因斷裂情況,可以克服以上困難,并且快速、易于操作,有助于BL的輔助診斷。
