The principle and scope of application of Escherichia coli 0157 test tablets
1, principle and scope of application
Escherichia coli 0157 test piece is a pre prepared, disposable medium product, containing selective media and specific enzyme chromogenic agent, and colony on the test piece is blue, which can shorten counting time and enhance counting effect. This product is suitable for the determination of Escherichia coli 0157:H7 in all kinds of food and drinking water.
2. Methods of use
2.1, sample treatment: take sample 25mL (g) into the sampling tank or homogenized cup containing 225mL sterilized phosphate buffer diluent (or normal saline), make the 1:10 sample homogenate, and adjust the sample homogenization pH to 6.6 to 7.2 when necessary, with 1mol/LNaOH or 1mol/L HCl solution. Sterilize the pipette with 1mL, absorb the 1:10 sample and homogenize 1mL, pour into the test tube containing 9mL diluent, shake it up and become the sample of 1:100, homogenize it, and so on, and make the sample such as 1:1000 dilution to homogenize, and change one suction tube at a time.
2.2, inoculation: general food should be selected by 2~3 dilution tests. Liquid samples containing less bacteria (such as drinking pure water and mineral water) can be directly extracted from the original liquid for testing. The test piece was placed on the flat test table, the upper membrane was uncovered, the 1mL sample was collected by aseptic pipette, and the sample was uniformly distributed to the paper. The top layer was slowly covered and placed at 10s for about two pieces. At the same time, a blank negative control was made.
2.3, c*tion: put the test pieces together and put them back in the original self sealed bag and seal them. The transparent face is placed in the incubator with constant temperature, and the number of stacked pieces is not more than 12 pieces. The temperature of general food culture is 36? 1 C and 15 ~ 24h, and the culture temperature of aquatic products is 30 and 1 C, and the culture is 48h.
3, result interpretation
After culture, the purple red colonies on the test tablets were Escherichia coli 0157:H7, and the blue colony was the colony of other coliform colony. Samples of positive colonies are best verified by other more reliable methods, and at least repeat samples of the samples without conditions.
