三，免疫組化步驟
1，  切片，烤片60℃，1h；
2，脫蠟及復(fù)水
   二甲苯10min，二甲苯10min，100％乙醇5min，95％乙醇5min，90％乙醇5min，85％乙醇5min，80％乙醇5min， 75％乙醇5min，60％乙醇5min，50％乙醇5min，30％乙醇5min，自來水1min，雙氧水1min；
3，1份30％H2O2加10份蒸餾水，室溫10min，蒸餾水洗3次，每次3min；
4，微波修復(fù)
將切片浸入0.01M枸櫞酸緩沖液，微波中zui大火力（98℃-100℃）加熱至沸騰，冷卻（約5-10min），反復(fù)兩次；
5，將切片自然冷卻至室溫，PBS洗滌3次，每次5min；
6，封閉，5％BSA，室溫20min，甩去多余液體；
7，滴加一抗，37℃，1h，或者4℃過夜；
8，PBS洗滌3次，每次3min；
9，滴加二抗，37℃，15-30min；
10，PBS洗滌3次，每次3min；
11，滴加SABC，37℃， 30min；
12，PBS洗滌3次，每次5min；
13，1ml蒸餾水中分別滴加顯色劑，混勻；
14，DAB顯色劑配置好后，滴加于切片，室溫，鏡下檢測反應(yīng)時(shí)間（約5min）；
15，自來水沖洗干凈，過蒸餾水；
16，蘇木素復(fù)染2min，自來水沖洗；
17，脫水
30％乙醇3min，50％乙醇3min，70％乙醇3min，80％乙醇3min，90％乙醇3min，95％乙醇3min，100％乙醇3min，二甲苯20min；
18，樹膠封片，鏡檢。

Third, immunohistochemistry steps
1, sliced, baked slices 60 ℃, 1h;
2, dewaxing and rehydration
   10% xylene 10min 100% ethanol 5min 95% ethanol 5min 90% ethanol 5min 85% ethanol 5min 80% ethanol 5min 75% ethanol 5min 60% ethanol 5min 50% ethanol 5min 30% Ethanol 5min, tap water 1min, hydrogen peroxide 1min;
3,1 30% H2O2 plus 10 parts of distilled water, room temperature 10min, distilled water 3 times, each 3min;
4, microwave repair
The slices were immersed in 0.01M citrate buffer, the maximum heat in the microwave (98 ℃ -100 ℃) was heated to boiling, cooling (about 5-10min), repeated twice;
5, the sections were cooled to room temperature, washed with PBS 3 times, each 5min;
6, closed, 5% BSA, room temperature 20min, throw off excess liquid;
7, dropping a primary antibody, 37 ℃, 1h, or overnight at 4 ℃;
8, washed with PBS three times, each time 3min;
9, dropping secondary antibody, 37 ℃, 15-30min;
10, PBS washed 3 times, each 3min;
11, dropping SABC, 37 ℃, 30min;
12, PBS washed 3 times, each 5min;
13,1 ml of distilled water were dropped reagent, mixing;
14, DAB reagent configuration, dropping in slices, room temperature, microscopic detection reaction time (about 5min);
15, tap water rinse, distilled water;
16, hematoxylin counterstain 2min, tap water rinse;
17, dehydration
30% ethanol 3min, 50% ethanol 3min, 70% ethanol 3min, 80% ethanol 3min, 90% ethanol 3min, 95% ethanol 3min, 100% ethanol 3min,
18, gum seal, microscopic examination.
