石蠟切片免疫組化染色步驟
1、載玻片的處理：
抗原修復(fù)過(guò)程中，由于高溫、高壓、輻射等諸多因素的影響，極易造成脫片。為保證試驗(yàn)的正常進(jìn)行，可選用我公司提供的ZLI-9001 APES、ZLI-9003 HistogripTM或ZLI-9005 Poly-L-Lysine 等幾種試劑，對(duì)已清洗的載玻片進(jìn)行處理。具體方法如下：
1.1 APES：現(xiàn)用現(xiàn)配。將洗凈的玻片放入以1:50 比例丙酮稀釋的APES 中，停留20~30 秒鐘，取出稍停片刻，再入純丙酮溶液或蒸餾水中涮去未結(jié)合的APES，置通風(fēng)櫥中晾干即可。用此載玻片撈片時(shí)應(yīng)注意組織要一步到位，并盡量減少氣泡的存在，以免影響染色結(jié)果。
1.2 HistogripTM：將洗凈的玻片放入以1:50比例丙酮稀釋的Histogrip液中，停留1~2 分鐘，然后用雙蒸水快速清洗三次，室溫干燥或60oC 烤箱烘烤一小時(shí)，裝盒備用。
1.3 Poly-L-Lysine：將洗凈、干燥的載玻片放入以1:10 比例去離子水稀釋的多聚賴(lài)氨酸溶液中，浸泡5 分鐘，60oC 烤箱烘烤一小時(shí)或室溫過(guò)夜干燥。裝盒備用。試驗(yàn)中使用的器具均為非玻璃制品。
2、常用酶消化：
2.1 胰蛋白酶：一般使用濃度為0.05%~0.1%，消化時(shí)間為37℃、10~40 分鐘，主要用于細(xì)胞內(nèi)抗原的顯示。
2.2 胃蛋白酶：一般使用濃度為0.4%，消化時(shí)間為37℃、30~180 分鐘，主要用于細(xì)胞間質(zhì)抗原的顯示，如：Laminin（層粘蛋白），Collagen IV（IV型膠原）等。
2.3 皂素（Saponin）：一般使用濃度為2~10?g/ml 的saponin 溶液，消化時(shí)間為室溫孵育30 分鐘。

1.3 Poly-L-Lysine: Place the washed and dried slide in a polylysine solution diluted 1:10 in deionized water, soak for 5 minutes, bake in a 60oC oven for one hour or overnight at room temperature . Cartoning spare. The test equipment used are non-glass products.
2, commonly used enzyme digestion:
2.1 trypsin: general use concentration of 0.05% to 0.1%, the digestion time of 37 ℃, 10 to 40 minutes, mainly for the display of intracellular antigens.
