實(shí)際上是一個(gè)連續(xù)的膜囊和膜管網(wǎng),可分為糙(粗)面內(nèi)質(zhì)網(wǎng)(RER, Rough Endolasmic Reticulum)和光(滑)面內(nèi)質(zhì)網(wǎng)(SER, Smooth Endolasmic Reticulum)兩大部分。蛋白質(zhì)合成編輯(一)蛋白質(zhì)合成蛋白質(zhì)都是在核糖體上合成的,并且起始于細(xì)胞質(zhì)基質(zhì),但是有些蛋白質(zhì)在合成開(kāi)始不久后便轉(zhuǎn)在內(nèi)質(zhì)網(wǎng)上合成,這些蛋白質(zhì)主要有:①向細(xì)胞外分泌的蛋白、如抗體、激素;②跨膜蛋白,并且決定膜蛋白在膜中的排列方式;③需要與其它細(xì)胞組合嚴(yán)格分開(kāi)的酶,如溶酶體的各種水解酶;④需要進(jìn)行修飾的蛋白,如糖蛋白。C. Milstein()發(fā)現(xiàn)從骨髓瘤細(xì)胞提取的免疫球蛋白分子N端要比分泌到細(xì)胞外的N端多出一段。G. Blobel和D. Sabatini等根據(jù)進(jìn)一步的實(shí)驗(yàn),提出了信號(hào)假說(shuō)(Signal hyothesis),認(rèn)為蛋白質(zhì)上的信號(hào)肽,指導(dǎo)蛋白質(zhì)轉(zhuǎn)至內(nèi)質(zhì)網(wǎng)上合成。Blobel因此項(xiàng)發(fā)現(xiàn)獲年諾貝爾生理醫(yī)學(xué)獎(jiǎng)。蛋白質(zhì)轉(zhuǎn)入內(nèi)質(zhì)網(wǎng)合成至少涉及種成分:①信號(hào)肽(signal etide),是引導(dǎo)新合成肽鏈轉(zhuǎn)移到內(nèi)質(zhì)網(wǎng)上的一段多肽,位于新合成肽鏈的N端,一般~個(gè)氨基酸殘基,含有-個(gè)帶正電荷的非極性氨基酸,由于信號(hào)肽又是引導(dǎo)肽鏈進(jìn)入內(nèi)質(zhì)網(wǎng)腔的一段序列,又稱開(kāi)始轉(zhuǎn)移序列(start transfer sequence)。②信號(hào)識(shí)別顆粒(signal recognition article,SR),由種結(jié)構(gòu)不同的多肽組成,結(jié)合一個(gè)S RNA,分子量KD,屬于一種核糖核蛋白(ribonucleorotein)。SR與信號(hào)序列結(jié)合,導(dǎo)致蛋白質(zhì)合成暫停。③ SR受體(SR recetor),是膜的整合蛋白,為異二聚體蛋白,存在于內(nèi)質(zhì)網(wǎng)上,可與SR特異結(jié)合。

Contains - a positively charged non-polar amino acid, also known as the start transfer sequence because the signal peptide is a sequence that guides the peptide chain into the lumen of the endoplasmic reticulum. 2 signal recognition particles (signal recognition article, SR), composed of different species of polypeptide composition, combined with a S RNA, molecular weight KD, belongs to a ribonucleoprotein (ribonucleorotein). SR binds to signal sequences and causes protein synthesis to pause. 3 The SR receptor, an integral protein of the membrane, is a heterodimeric protein that is present on the endoplasmic reticulum and can specifically bind to SR.
